Title: Improvement of clinical islet transplant outcome by enhancing islet qualityand immunoisolating islets

Abstract

For clinical islet transplantation, harvesting functional islets and maintaining long-term graft survival post-transplantation are critically important and challenging. To recover high quality of human islets, our efforts have focused on examining donor characteristics, refining islet isolation procedure, and understanding the effect of marijuana use and islet health, as well as enhancing islet function by simultaneously isolating intra-pancreatic tissue-derived MSC and islets. To achieve stable graft function and attenuate toxic effect of the systematic immunosuppressive medications, we targeted on immunoisolation of pancreatic islets by encapsulation. Despite promising results on encapsulated islet transplantation in various animal models, the field has failed to apply this technology in well-documented clinical trials. The clinical application of encapsulation technology is limited by raw material variability and species-specific differences in the host response to encapsulated islet grafts. This seminar will address recent findings on enhancing islet quality prior to transplantation, as well as some results of encapsulation study in rodent and pre-clinical non-human primate models.
