講演要旨

Hair follicle stem cells cluster in a specialized region of the hair follicle called the bulge, and express a number of transcription factors which regulate their function. We found that the transcription factor Runx1, controls hair follicle stem cell activation, as well as the initiation and maintenance of epithelial skin tumors. Recently, we uncovered a role of Runx1 from within the epithelial stem cells to promote transcription of target genes and eventually secretion of molecules that remodel components of the stem cell environment, or the niche. This included remodeling of vasculature arrangements around the hair follicle during the hair adult homeostatic cycle. Next, we examined if in turn blood vessel arrangements in the skin have a role in hair cycling and stem cell activation. Thus, we have employed blood vessel mutants in which the skin vasculature arrangement around the adult hair follicle is perturbed from within the endothelial cells and observed defects in stem cell activity and hair cycle. Our preliminary data suggest that BMP signaling might mediate the vasculature effect on hair follicle stem cells. Our work demonstrates that vasculature may play a niche role for hair follicle stem cells in mouse skin. Importantly, we also find that the epithelial stem cells remodel their niche vasculature to create the proper 3D arrangements in preparation for a new growth cycle. These data demonstrate a coordinated two-directional cross-talk between stem cells and the endothelial niche.                     (言語:英語 / Language English)
