講演要旨

Genome-wide transcriptome studies have shown that mammalian genomes are pervasively transcribed, generating a wide variety of transcript isoforms. Most transcripts do not code for proteins, but may have regulatory roles as non-coding RNAs. To discover and analyze coding and non-coding gene regulation, our laboratory takes advantage of Cap Analysis Gene Expression (CAGE) transcriptome profiling technology, which was developed at RIKEN and is heavily used in the FANTOM projects. CAGE detects and quantifies transcription initiation at single-nucleotide resolution, allowing the discovery and analysis of promoters and enhancers on a genome-wide scale. By integrating CAGE with Hi-C chromatin conformation data, we associate enhancers with the promoters they regulate, and assign functions to non-coding RNAs based on their physical association with coding genes. More recently, we apply our expertise in basic genomics to biomedical problems such as the behavior of cells upon infection by SARS-CoV-2, and the patient-specific drug response in personalized medicine for the treatment of acute myeloid leukemia. Complementary to sequencing-based approaches, we are currently developing novel techniques to image specific chromatin regions using electron microscopy. (Language:English)
