講演要旨

Bunya- and orthomyxovirus families including influenza A virus (IAV) snatch 5’
cap portion of host RNAs for their replication. Here we report the specific
involvement of host 2’O-ribose methyltransferase MTr1 in the replication of IAV
and influenza B virus (IBV) among varieties of cap snatching viruses. We
identified tri-fluoromethyl-tubercidin (TFMT) as an MTr1 inhibitor with the
antiviral specificity to IAV and IBV by in silico screening of chemical compounds
for the S-adenosyl-L-methionine binding pocket of MTr1, together with cellbased
antiviral screening. MTr1 inhibition impairs the association of host cap
RNAs with viral polymerase. TFMT notably exhibited synergy with existing
drugs, efficacy in human lung explants and in vivo in mice. Our work will provide
new insight into host-directed therapeutic modality against influenza viruses.
