演題: Quantitative behavior of CDK activity in living cells

講演要旨

Cyclin-dependent kinase (CDK) plays an essential role in determining the
temporal ordering of the cell cycle phases. However, despite significant
progress in studying regulators of CDK, it remains elusive how they
coordinately affect CDK activity at the single-cell level and how CDK
controls the temporal order of cell cycle events. This could be due to the
lack of tools to monitor CDK activity in living cells. Recently, we developed
a biosensor to visualize the dynamics of CDK activity in living fission yeast
and mammalian cells, named Eevee-spCDK. In this seminar, we decipher
the quantitative behavior of CDK activity in several selected mutants, by
using Eevee-spCDK. We show that although the cyclin levels at G2/M
transition differ among the mutants, CDK activity converges to the same
level among them. We discuss the obtained data in the context of the
previously proposed models depicting the mechanism to progress the cell
cycle properly.(Language: English)
